You didn't mention whether you can get in the queue for directory requests.
Doppler should not be a problem for the uplink, you don't need to adjust it. 
Just use the standard uplink frequency, the 2M receivers on the sats are 
wide enough to handle the Doppler.
The most common problems I have found to be 1) incorrect audio levels from 
TNC into the transciever, this is an important setting;
2) deviation for the transmitter set to 3 kHz, as opposed to the standard 5 
kHz.

If you don't have access to a station monitor, I recommend finding someone 
locally with a TH-D7A or TM-D700.  Put these into 9600 bps packet mode and 
send files to them.  Then adjust your input level to the transceiver for 
maximum throughput.
